RECONSTITUTION PROCEDURE

All Ares Research CJC-1295 vials arrive as lyophilized powder sealed under inert gas. Reconstitution requires bacteriostatic water to prevent bacterial contamination. Do not use tap water, sterile water, or saline alone.

STEP 1

Temperature Equilibration

Remove vial from -20°C storage and allow 15 minutes at room temperature before reconstitution.

STEP 2

Volume Calculation

For a 10mg vial, inject 10ml bacteriostatic water to achieve 1.0 mg/ml. Use a sterile insulin-grade syringe for accurate measurement.

STEP 3

Slow Injection

Insert needle at 45° angle through rubber stopper. Inject solvent gently along the interior glass wall. Do not inject directly onto the powder cake.

STEP 4

Gentle Agitation

Roll vial between palms for 60 seconds. CJC-1295 dissolves readily. Visual inspection: solution should clarify completely.

Reconstituted CJC-1295 is stable for 7 days at 2–8°C. For long-term storage, aliquot into separate vials and freeze at -20°C immediately. Avoid repeated freeze-thaw cycles.

CONCENTRATION & DOSAGE REFERENCE

Standard Ares Research CJC-1295 vials contain 10mg of lyophilized GHRH analog. The table below shows concentration after reconstitution.

Vial Content10 mg
Bacteriostatic Water10.0 ml
Concentration1.0 mg/ml (1,000 mcg/ml)
0.1 ml (10 units)100 mcg
0.05 ml (5 units)50 mcg

RESEARCH DOSAGE RANGES

  • Somatotroph GH secretion (GH3 / primary pituitary): 0.1–10 mcg/ml in serum-free DMEM/F12, 1–24 hours
  • Hepatocyte IGF-1 upregulation: 0.1–1.0 mcg/ml in HepG2 or primary hepatocyte cultures, 24–72 hours
  • Adipocyte lipolysis assays: 0.01–1.0 mcg/ml in differentiated 3T3-L1 cultures, 6–24 hours
  • DPP-IV resistance validation: pre-incubate with recombinant DPP-IV (50–100 ng/ml) for 2 hours, then assay GH release

STORAGE & STABILITY

CJC-1295 is highly stable in lyophilized form due to its compact 29-aa structure and lack of oxidation-sensitive residues.

  • Lyophilized powder: stable ≥ 36 months at -20°C, protected from light
  • Reconstituted solution: 7 days at 2–8°C; aliquot and freeze at -20°C for long-term storage
  • Aliquots at -20°C: stable ≥ 6 months; thaw each aliquot once and discard after use
  • Protect from UV and fluorescent light exposure at all stages
  • pH of reconstituted solution should remain 6.5–7.5; discard if discoloration or precipitation occurs

— FREQUENTLY ASKED QUESTIONS

Q: What water should I use to reconstitute CJC-1295?

Use bacteriostatic water (0.9% benzyl alcohol) exclusively. Sterile water lacks a preservative and allows bacterial growth. Never use tap water, distilled water, or saline without bacteriostatic additives.

Q: How long does reconstituted CJC-1295 last?

Reconstituted CJC-1295 remains stable for approximately 7 days when stored at 2–8°C in the dark. For longer storage, aliquot into separate sterile vials and freeze at -20°C. Avoid freeze-thaw cycling.

Q: What concentration should I use for GH secretion assays?

For GH3 rat pituitary somatotrophs or primary anterior pituitary cell cultures, CJC-1295 is typically evaluated at 0.1–10 mcg/ml in serum-free DMEM/F12. GH secretion is measured by radioimmunoassay or ELISA at 1, 3, 6, and 24 hours. Always include a vehicle control.

Q: Does the DAC modification affect reconstitution?

The DAC (Drug Affinity Complex) is a C-terminal modification that does not significantly alter solubility or reconstitution behavior. CJC-1295 dissolves readily in bacteriostatic water within 60 seconds. The DAC group becomes biologically active only after intravenous or subcutaneous administration in in-vivo models.

Q: Is CJC-1295 the same as Tesamorelin?

No. CJC-1295 is a 29-amino-acid truncated GHRH analog with DAC modification and DPP-IV resistance. Tesamorelin is a 44-amino-acid full-length GHRH analog with trans-3-hexenoic acid conjugation. They differ in sequence length, half-life, stability mechanism, and research applications.
